General Objective: Optimize an electrochemical sensor for the detection of hydrogen peroxide (oxidative burst) resulting from stress by Cd(II) ions, in stem and leaf stem cells of Pennisetum purpureum Schumach and Pennisetum violaceum grown in vitro as a model device for selection. of plants useful in phytoremediation.
Specific objectives:
- Optimize a method using electrochemical techniques for the quantification of peroxide in standard solutions by obtaining figures of merit.
- Obtain cell suspensions of stem and leaves of Pennisetum purpureum Schumach as explants for in vitro culture.
- Obtain cell suspensions of Pennisetum violaceum stem and leaves as explants for in vitro culture.
- Quantify hydrogen peroxide by chronoamperometry as a signal of cadmium accumulation in cell suspensions of Pennisetum purpureum Schumach exposed to cytotoxic levels of this metal.
- Quantify hydrogen peroxide by chronoamperometry as a signal of cadmium accumulation in cell suspensions of stem and leaves of Pennisetum violaceum exposed to cytotoxic levels of this metal.
- To quantify the accumulation of cadmium in stem and leaf cell suspensions of Pennisetum purpureum Schumach and Pennisetum violaceum exposed to cytotoxic levels of this metal by atomic absorption spectroscopy.
- Selection of the plant with the best attributes to be applied in phytoremediation.
